Will this seal a seam in plywood on a roof before doing the underlayment and shingles


This can be used for that purpose. However, our Roof Deck Seam Tape was made for that purpose. It is also carried by Lowe's - Sku #494015. I'm not sure what part of the country you are in so you should check the application temperatures on the label.
