Does holes have to be predrilled to use this product in the wood or concrete slab

Yes, if you are using it to hold a piece of 2x I would suggest you pretrial your holes on the 2x and use it as a guide to locate the hole you are drilling into concrete, Then finish drilling with the right bore masonry bit. Blow out the hole of excess dust and add some construction adhesive for additional hold.

Concrete screws are self tapping. And I would not use them in wood, they say concrete for a reason.
