Can this be used with a no dig mailbox spike, or will there not be enough clearance to fit the plastic around it?


Only if you are able to bury the spike low enough below ground that the mailbox post is only sliding over the 4 x 4 wood (to reach ground level). This mailbox post will not slide over the metal spike portion.
